Abstract

A major problem in the confocal microscopy of thick biological tissue is that the resolution deteriorates and the signal level falls rapidly as the microscope is focussed into the specimen. This is unavoidable and is caused by refractive index mismatches. We intend to develop a system which both measures the wavefront aberration and uses active feedback to a phase correction element. This results in near diffraction limited imaging throughout the whole volume. The key element is a novel design of wavefront sensor with a particularly high signal to noise ratio and which also exhibits confocal sectioning. The system will be used to image biologically important tissue in collaboration with the Department of Pharmacology at Oxford.
