Abstract

We propose to exploit the use of mass spectrometry in the study of protein - ligand interactions with the overall objective of evaluating the technique as a potential screening method for selecting ligands. This work will involve an in-depth study of the different factors stabilising protein - ligand interactions in solution and in the gas phase of the mass spectrometer. Using protein - ligand systems which are well characterised in solution we will assess the overall sensitivity of the mass spectrometry method, the quality and rapidity of results and pitfalls that may arise in interpretation of data. Furthermore, we intend to examine the conditions which allow us to observe water molecules in the binding site of the protein and, using hydrogen exchange methods, the conformational changes which occur upon ligand binding.
