Abstract
1) The genes encoding alkene mono-oxygenase (AMO) from Xanthobacter Py2 will be sequenced and over-expressed in Pseudomonas species and E. coli. 2) The amo A,B and C genes from Mycobacterium L1 will be cloned and sequenced, and the homology between these sequences and those from Mycobacterium M156 and Nocardia corallina and B276 established. 3) Intramolecular hybrids based on regions of sequence divergence between A and C genes from the three organisms cited in (2) will be created to discover the regions which have the greatest effect on substrate and product specificity. 4) Specific regions will then be analysed in more detail.
