Abstract

Recent demonstrations of the value and applications of a sensitive fluorogenic microscope-based assay of beta-galactosidase (lacZ) which detects activity at the single cell level has led the applicant to address current limitations to this approach at a generic level. The potential contribution of differences in substrate penetration to the site of hydrolysis will be approached by synthesis and evaluation of novel substrates which emit two distinct signals in proportion to local substrate and reaction product concentrations; distinction between recent and persisting reporter molecules will be improved by development of labile lacZ mutants; and the capacity of the microscope-based assay to analyse large populations of cells will be enhanced by image analysis software development.
