Abstract

We have used 2D gels and phage biopanning to identify immunogens which are seen during murine infection with Salmonella typhimurium. We now propose to clone, overexpress and affinity purify a selection of these proteins and also, known adhesins (approx. 100 proteins) and, to evaluate their immunological importance in the humoral and cell mediated immunity which arises during infection of mice and calves. Using inbred/congenic mice, the importance of host genetic background on polypeptide recognition will also be addressed and the potential of a selection of the antigens to elicit protective immunity against virulence challenge will be examined. The study will provide evidence for the value of the antigens as potential vaccine components.
