Abstract

In our previous study in which we investigated the effects of fish oil fatty acids on the expression of lipoprotein lipase in human adipose tissue we made the unexpected and novel observation that fish-oil induced increases in plasma LDL-C were seen only in subjects with an apoE4 genotype (approx 20 per cent population). The present study will employ 2H-leucine labelled apoB-100 and 2H-glycerol labelled TG turnover studies to examine the underlying mechanisms involved. The hypothesis is that increased LDL-C in apoE4 subjects is the result of a greater impact of the docosahexaenoic acid fraction of fish oils on hepatic and extra hepatic metabolism resulting in: (i) increased fractional production rate (FPR) of LDL from VLDL and (ii) decreased fractional catabolic rate (FCR) of LDL particles by the hepatic LDL receptor.
