Abstract

Obesity and infertility are symptoms of inappropriate body weight. Since both axes are regulated by nutritional feedback within the same brain region (hypothalamus), they share common mechanisms. Better knowledge of these is needed to develop correct therapies for sustained weight loss and improved reproductive health. Nutrition influences reproduction via gonadotrophin releasing-hormone (GnRH) output. Our animal model shows that the hypothalamus has a nutritional memory, dissociating dynamic and static feedback, with differential effects on GnRH. We hypothesise that peripheral feedback from leptin, insulin and ghrelin modulates GnRH through hypothalamic pathways shared with energy balance regulation; these will be tested in our animal model using in vivo intrahypothalamic sampling:infusion and in vitro gene expression analysis.
