Abstract
This proposal combines innovations in the molecular biology of wheat proteins with developments in spectroscopic methods to understand the way in which the sequence details of the repeat peptides of the high molecular weight subunits of gluten affect their behaviour. The behaviour of repeat peptides with perfect sequences will be compared with those having controlled mutations using FTIR and NMR spectroscopy methods, both statically and under mechanical deformation. Results will be interpreted in terms of the loop and train model for high molecular weight subunits and used to predict the way in which sequence affects behaviour.
