Abstract

Conventional cDNA libraries are invaluable for genome research as they provide access to functional sequences in a compact format. Nevertheless inefficiencies in their isolation are known e.g. the representation of sequences is proportional to the degree of transcription in the tissue of origin. Thus rarely expressed sequences are underrepresented and this is only partially circumvented by normalisation. Knowledge of map location is critical to narrowing the search for a gene however sequencing the entire genome or even millions of ESTs is not currently an option for farm animals. Thus strategies for focusing on regions of interest are essential. We therefore propose to combine the strengths of a cDNA strategy with an approach that includes map information to generate and evaluate resources (i.e. region specific normalised cDNA libraries) for gene discovery in farm animals.
