Abstract

The aim this work is to capitalise on a number of recent scientific advances, combining them to develop an effective gene targeting system for plants. (1) Promising negative selection systems already shown to work in plants will be adapted for gene targeting. (2) Bacterial recombinases shown to enhance recombination when introduced singly into plants, will be co-ordinately expressed in plants along with other potential recombination- enhancing proteins. The plants transformed will already contain an artificial recombination substrate so that changes in somatic and meiotic recombination frequencies can be easily assessed. Finally, the best results from these two approaches will be combined to develop a gene targeting system for plants.
