Abstract

The feasibility of manipulating cell wall polysaccharide synthesis through alteration of Golgi sugar nucleotide transporter activity will be demonstrated. We will confirm our identification of a gene from Arabidopsis encoding a protein that translocates GDP-mannose into the Golgi lumen, using yeast complementation, and localisation of the protein in plants by microscopy. Next, we will demonstrate the role of the transporter in GDP-mannose transport, by use of mutant plants and of a transport assay with isolated plant Golgi vesicles. Finally, we will investigate the effect on polysaccharide synthesis of the manipulation of Golgi luminal GDP- mannose availability through selection and generation of transporter mutants and over- expressing plants. This work will thus provide enabling technology for the commercial generation or selection of plants with altered cell wall polysaccharides.
