Abstract

Enterohaemorrhagic E. coli [EHEC] infections are an increasingly important zoonosis. Putative virulence factors have been identified, but their role in pathogenesis and host specificity remains unclear. We aim to study the interaction of wild type and defined mutant strains of EHEC with intestinal mucosae from humans and cattle, natural host species. Early host/pathogen interactions will be assessed in primary intestinal biopsies using in vitro organ culture. Subsequent events in pathogenesis will be studied using the ligated intestinal loop model in cattle which allows quantification of intestinal invasion, secretory and inflammatory responses. Translocation of specific virulence factors will be assessed by immunofluorescent and confocal microscopy. Signature tagged mutagenesis will be exploited in vivo to identify novel virulence genes.
