Abstract

Regulation of nutrients of gastrointestinal gene expression is important in the response of the gut to diet. An integral understanding of adaptation to diet requires the study of coordinated gene responses. The aim of this project is to investigate the hypothesis that the adaptive response to dietary protein levels involves the coordinated regulation of peptide and amino acid transporters, by elucidating patterns of coordinate regulation of gastrointestinal amino acid and peptide transporters, at the molecular level, in response to total dietary nitrogen levels and to specific amino acids. Levels of transporter expression in human intestinal Caco-2 cells will be quantified by RT-PCR, immunohistochemistry and western blotting. The functional consequences of regulation will be established by studies on the cellular uptake and transport of radiolabelled substrates. Comparative studies in the rate will place the findings in a physiological context. The results will be relevant to a number of dietary health issues.
