Abstract

We previously reported the isolation of ES lines from refractory murine genotypes by selective ablation of differentiated cells, demonstrating that a functional ES lineage does exist in these strains. The technique involved explanting embryos from transgenic mice under selection for the expression of an engineered ES-specific selectable marker. Here we seek funding (1) to test the strategy in existing lines of transgenic pig which contain the same transgene, and (2) to further develop the technology for use with embryos derived from non-transgenic donors by direct selection of transfected embryos (transient ablation). Cloning of constructs and characterisation of porcine transgene expression will be done in year 1. Direct selection of transfected murine embryos will be assessed in years 1 and 2, and analogous porcine experiments undertaken in year 3.
