Abstract

Entry into stationary phase, mediated in large part by the RpoS sigma factor, greatly enhances the resistance of Salmonella species to a variety of stresses, including starvation, hyperosmosis, oxidation, heat and low pH. Currently, however, less than 50% of the proteins induced in early stationary phase have been identified. To achieve a comprehensive analysis of such proteins in Salmonella typhimurium, high resolution two-dimensional gel electrophoresis and microsequencing will be employed. Genes encoding representative novel stationary phase-regulated proteins will be cloned and specifically mutated to assess their contribution to Salmonella survival in various food matrices.
