Abstract

Fruit and vegetable quality is affected significantly by the textural properties of tissue primary cell walls [PCW]. Pectin, an important polysaccharide in PCW of most fruit and vegetable crops, compromises several structural domains, including homogalacturonan [HG] - a (1-4)-alpha-linked galacturonan with varying degrees of methyl esterification [DME] on carboxyls. Pectin DME is known to be important in determining fruit and vegetable quality. The aims are to investigate the regulation of DME during pectin biosynthesis, and to attempt the full molecular characterisation of pectin methyltransferase, the key enzyme responsible for forming the methyl ester linkages.
