Abstract
Our overall aim is to elucidate the molecular details, intracellular events and mechanisms of regulation of the assembly and secretion of chylomicrons by enterocytes. Based on our findings we will investigate how these events are modulated by diets of different lipid composition. These studies will use rabbits and hamsters as experimental animals and results will be confirmed as far as is practically possible using fresh human small intestine mucosa. Our experimental approach will be to isolate enterocytes from animals fed different diets and using pulse- labelling combined with cell fractionation and biochemical analyses follow the intracellular transit of apo-B48 and its assembly with chylomicron lipids.
