Abstract
The aims are to investigate the parameters that control the efficiency of expression and maintenance of genes transformed into land plant plastids. This will involve: (1) the construction of minimal expression cassettes with reporter and marker genes; (2) modification of minimal cassettes to promote:- site-specific targeting, plasmid amplification and maintenance of free plasmids in plastids; (3) transient expression assays in isolated chloroplasts and cells; (4) production of stable chloroplast transformants; (5) analysis of the structure, mitotic and meiotic stability, and expression of plastid transgenes.
