Abstract

This project aims to test the hypothesis that the spatial and temporal patterns of expression and regulation of the gene products responsible for the absorption of intact peptides from the gut lumen to the blood are controlled by nutrients. Reagents for in situ and antibody studies will be generated. Spatial (duodenal-to-colonic axis) and temporal (crypt-to-villus axis) patterns of peptide transporter mRNA and protein expression will be investigated by in situ hybridisation and by immunohistochemistry in the human small intestine. Human intestinal Caco-2 cells will be used as an in vitro model to study temporal patterns of expression, the role of nutrients on expression (with comparative experiments in mice in vivo) and to delineate possible second messenger systems involved in regulation of peptide transporter gene expression and activity.
