Abstract

The discovery that chemical disruption of aromatic amino acid biosynthesis can lead to plant death has made this metabolic `shikimate' pathway an attractive target for herbicide development. We have recently obtained crystals of 3-dehydroquinate synthase (DHQS), an essential enzyme in the shikimate pathway, as a complex with a herbicidal compound that diffracts to 1.8 angstroms. We propose to establish a high resolution crystal structure of this enzyme-inhibitor complex. This is expected to lead to a hypothesis about the mode of binding of the inhibitor and substrate; indicate the chemistry of the catalytic mechanism; and aid the design of other useful compounds. In parallel, we will express and purify a bi-functional DHQS/EPSPS enzyme.
