Abstract

The project exploits the application of molecular techniques for the quantification of fungi in soil in order to analyse the epidemiological effectiveness of a hyperparasite as a biocontrol agent. Specifically the objectives are: 1) to apply previously developed and tested monoclonal antibody (mAb) based immunological assays with beta-glucuronidase markers to quantify population cycles of Rhizoctonia solani and Trichoderma harzianum and to determine the relative epidemiological roles of components of hyperparasite biomass in biological control of the pathogen; 2) to use mathematical models to synthesise the population dynamics of R. solani and T. harzianum, obtained from the mAb-based assays, in order to identify critical points in the interaction that would optimise the effectiveness of biocontrol; 3) to use mAb-based assays to analyse the effects and interaction of T. harzianum on the local multiplication of R. solani in the rhizosphere and bulk soil; 4) to continue quantification of R. solani densities in naturally infested soils, using molecular techniques.
