Abstract
The absorption of intestinal sugars is known to involve multiple steps and at least three transporter proteins, GLUT2 (a glucose/fructose transporter), GLUT5 (a fructose transporter) and the sodium dependent transporter SLGT1. To assess the separate transport contributions and specificities of these proteins we plan to express these proteins in Xenopus oocytes. We will develop new bis-hexose probes for interacting with GLUT5 (bis-fructose analogues) and SLGT1 (bis-glycoside analogues). These probes will be used to investigate conformational changes during transport catalysis. The selectivity of the transporters for fructose vs. glucose will be investigated by constructing chimeric transporters from GLUT5 (specific for glucose) and GLUT3 (specific for glucose). These chimeric proteins will be studied using hexose analogues and photo- affinity labels. The photo-affinity labels will also be used to study changes in transporter targeting during development and under conditions of different dietary manipulations. Taken together information about the kinetics, substrate selectivity and protein targeting/expression studies will greatly enhance our understanding of the mechanism and regulation of enterocyte sugar transport. This study is a joint venture linking the molecular approaches of the Glasgow group with the experience in photo-label/targeting of the Bath group.
