Abstract

1. By the sequencing, transcription analysis and targeted disruption of the (already cloned) whiA, H and J genes, we will complete the basic characterisation of a set of five known regulatory loci that together activate cytokinesis of aerial hyphae, leading to sporulation. 2. By SI mapping in defined null mutants transcriptional interdependence among the five genes will be evaluated. 3. By overexpression of E. coli appropriate whi gene products will be purified to raise antibodies (and for use in in vitro studies of DNA-protein (or protein-protein) interactions). 4. The effects of ectopic expression of the same whi genes in S. coelicolor will be evaluated, by Western blotting, S1 mapping and phenotypic analysis, to test in vivo the mechanistic hypotheses arising from 1-2.
