Abstract

Eight Ccm proteins catalyse the attachment of heme to polypeptide so as to generate a c-type cytochrome in plant mitochondria and many Gram negative bacteria. We will build on our recent success in achieving some of the Ccm-mediated steps in vitro by introducing new in vitro and in vivo strategies to elucidate the mechanism of the Ccm system in E. coli. Particular emphasis will be placed on heme provision, interactions between the Ccm proteins and apocytochromes, as well as studies of disulfide formation and reduction in the CXXCH motif of apocytochromes.
