Abstract

We have recently identified the new P-Rex family of guanine-nucleotide exchange factors, comprising P-Rex1 and its predicted homologue P-Rex2. P.Rex1 is a direct activator of the monomeric GTPase Rac, a key regulator of a huge range of cellular functions. P-Rex1 activity is regulated by a unique mechanism, synergistically by the lipid second messenger PtdIns(3,4,5)P3 and the betagamma subunits of heterotrimeric G proteins. It is required for G protein-dependant reactive oxygen species formation in neutrophil-like cells. We propose: i) to clone and characterise P-Rex2; ii) to study the phenotype of the P-Rex1 mouse we are currently generating; iii) to make and use a panel of P-Rex mutants to study in detail the mechanisms of P-Rex regulation.
