Abstract
Plant cells do not have microtubule organising centres. Without markers for MT nucleation we do not know how MT arrays are built, especially the cortical array essential for cell elongation. Promising pilot studies show that GFP-EB1 labels bodies at spindle poles (the classical location for MTOCs) and punctae on cortical MTs. Next we propose to clone the small multigene family. Labelling EB1 on plasma membrane disks with immunogold will allow us to visualise the MT nucleation sites and establish the orientation of cortical MTs. EB1 is known to form a complex and we will identify binding partners by affinity tags and proteomics. The developmental role of EB1 will be assayed by RNAi and by screening insertional mutant lines.
