Abstract

Homologous Recombination (HR) is vital for DNA repair and the generation of genetic diversity. Archaeal HR is poorly understood but appears related to eucaryal HR. We have identified and cloned several proteins implicated in archaeal HR, including the Holliday junction resolving enzymes Hjc and Hje. We will continue our characterisation of their enzyme mechanisms and substrate recognition, and examine the protein:protein and protein:nucleic acid interactions using a variety of biophysical, biochemical and proteomic technologies. Our aim is to gain an understanding of this important pathway at a molecular level. The project builds on a great deal of previous work and a strong set of collaborative partnerships.
