Abstract
The Brn-3a transcription factor activates specific genes in neurones and modulates neuronal differentiation and survival. Several of these effects such as activation of Bcl-2 and inhibition of apoptosis require an N-terminal activation domain which is present only in the longer of the two forms of Brn-3a. This N-terminal domain interacts with the Rin member of the Ras super-family. Moreover, Rin enhances target promoter activation by the long form of Brn-3a but has no effect on the short form. We will characterise the protein-protein interaction between Brn-3a and Rin, analyse the mechanisms by which Rin modulates transcriptional activation by Brn-3a and determine the effect of Rin on the biological activity of Brn-3a.
