Abstract

In contrast to excitatory glutamatergic synapses there is a paucity of molecular information regarding the composition, architecture and regulation of inhibitory synapses. My laboratory has discovered and cloned a novel 913 aa protein, GRIF-1, GABA-A Receptor Interacting Factor-1, by virtue of its specific association with GABA-A receptor beta2 subunits. Experimental and bioinformatics information support a role for GRIF-1 as a trafficking molecule in excitable tissues. This proposal aims to delineate the function of this novel gene by identifying associated proteins, refining the subcellular distribution of GRIF-1 and studying the effects of GRIF-1 on GABA-A receptor function, assembly, receptor internalisation, and cell surface expression.
