Abstract

This proposal aims to establish whether a biogenetic connection exists between the endoplasmic reticulum and peroxisomes in higher plants. Using the peroxisomal membrane protein Pex2p tagged with fluorescent proteins we will test whether trafficking is via the ER by using a combination glycosylation assays and pulse chasing with photoactivatable fluorescent proteins. Expression of mutant regulatory proteins in combination with a photobleaching strategy will test whether ER exit is COPII dependent. If not, a screen will be undertaken to search for mutants that mislocalise Pex2p to the ER or cytoplasm to identify components of this novel pathway. If trafficking proceeds by know pathways we will dissect the targeting signal of Pex2p instead. (Joint with grant 19029).
