Abstract

We will characterise the cytoskeletal functions of two S. cerevisiae genes, YPR171w and SYP1, identified as suppressor of the cytokinesis defective iqg1-1 mutation. Using a combination of genetic, biochemical and microscopy techniques we will define the role of these genes during cytokinesis and organisation of the cortical actin cytoskeleton. The phenotype of null mutations and possible synthetic interactions will be carefully examined with respect to cytoskeletal function. Protein and protein interactions and the localisation pattern of the two proteins will be monitored with particular emphasis on cell cycle changes and dependency relationships between Ypr171w, Syp1 and other previously characterised proteins; Iqg1, Hof1, Sla1, Rvs167.
