Abstract

Acute inflammatory conditions promote mast cells to release various inflammatory mediators. The process is promoted through cross- linking of the Immunoglobulin E (IgE) receptor and activation of intracellular signalling cascades. We have shown previously that antigen stimulation of mast cells results in activation of phospholipase D and that inhibition of phospholipase D inhibits mediator secretion. Many PLD functions can be ascribed to the promotion of remodelling of the actin cytoskeleton. We have recently identified a novel receptor-dependent association of PLD1 with F-actin in mast cells and this project is focussed on determining how this process is regulated and the physiological consequences.
