Abstract

NF-kB transcription factor is required for regulated immune, inflammatory and survival responses, while inappropriate activation of NF-kB is associated with inflammatory disease and cancer. Events which lead to release of NF-kB into the nucleus are well documented, but those determining transcriptional activity and specificity have yet to be determined. Objectives of this work are to: identify cellular proteins associated with the p65 subunit of NF-kB after it enters the nucleus; define the role of these associated proteins in vivo; compare p65 interacting proteins under a variety of activating conditions and in the presence of inhibitors; establish which promoters are bound by p65 when NF-kB signalling is induced by different stimuli
