Abstract

A major outstanding question in the CD45 tyrosine phosphatase research field is to understand the molecular actions of CD45 in regulating mature peripheral non-transformed lymphocytes. The aim of this project is to generate transgenic mouse lines using cre:loxP technology enabling conditional lineage-specific ablation of the CD45 gene in both the T and B lineages. Lymphocyte development will be normal in the mouse system as CD45 will be present during lymphoid maturation. Following CD45 gene ablation, immune responses, lymphocyte proliferation, cytokine secretion and cell survival will be investigated and correlated with signal transduction analysis of antigen receptors and their key CD28:CTLA-4 and CD19:CD22 co-receptors in T and B cells.
