Abstract

SK-1 is a novel rat protein. An hydropathy plot of the SK-1 sequence suggests that this protein has a transmembrane domain and a GPI-anchor. There is no convincing precedent for a mammalian protein with such a topology. Data from preliminary experiments on SK-1 are consistent with it having this topology. A GPI anchor has been shown to act as an apical targeting signal in polarised cells as has the addition of N- linked glycans. SK-1 possesses two potential sites for N-linked glycosylation (both apparently used). The aims of this project is are to confirm the predicted topology of SK-1, to investigate the intracellular trafficking of SK-1 and to begin to address the possibility that SK-1 might play a role in the apical delivery of proteins.
