Abstract

I recently identified in the amitochondrial protist Entamoeba histolytica a novel organelle of endosymbiotic origin, the mitosome. Phylogenetic and biochemical analyses suggest that the mitosome is related to hydrogenosomes and mitochondria but its physiological functions remain unknown. Using biochemical, gene cloning, and immunodetection methodologies this proposal aims to further characterise the mitosome by identifying additional enzymatic/structural components (which may reveal its physiological functions) and by analysing its morphology and fine structure. A mitosome purification protocol will be implemented and the protein import competence of purified organelles will be assessed using an in vitro protein import assay.
