Abstract
mRNA trafficking is a novel targeting mechanism of potential importance in subcellular organisation. Our recent evidence indicates that perinuclear mRNA localisation influences some nuclear-cytoplasmic protein trafficking. The aim of this project is to define the protein-RNA interactions that lead to perinuclear mRNA localisation. A protein binding to the 86nt localisation signal in the 3' untranslated region of c-myc mRNA will be isolated using RNA affinity methods. Having identified the protein, the gene will be cloned using a PCR approach. Recombinant protein will be made in bacteria and RNA-binding domains defined by UV cross-linking and sequencing. Antibodies will be raised and immunological methods, combined with RTPCR and gel shift assays, used to study the subcellular distribution of the protein and its interactions with RNAs and the cytoskeleton.
