Abstract
Activation of the class IA phosphoinositide-3 kinases (P13Ks) is of fundamental importance in controlling IL-3-induced proliferation. The aim of this study is to investigate the molecular events important for controlling this functional response and to specifically focus on: 1) the role of MEK1/MAPKs in mediating P13K-dependent signals; and 2) identification of other targets of P13K which are involved in regulating IL-3 proliferation. We will use transfectants we have generated to express Deltap85, wild-type p85 or p85 SH2 domains using the tetracycline-regulated expression system and LY294002 treatment to examine the following: functional role of MEK1/MAPKs; cell cycle status; pocket protein phosphorylation; expression and activity of cyclins, CDKs and CKIs; E2F activity and protein synthesis, using cellular, molecular and biochemical techniques.
