Abstract
This project aims to functionally characterise a nucleoside transporter from trypanosomes which is homologous to equilibrative nucleoside transporters in mammals. The transporter, termed P2, has been selected for study since it also carries trypanocidal drugs. Resistance to these drugs arises when the transporter is altered so that it no longer accumulates drugs to toxic levels. Mutations have already been identified in the gene encoding this transporter in one drug resistant line. We have several other drug-resistant lines and plan to identify the nature of mutations in these parasites too. The functional impact of each mutation will be identified by site-directed mutagenesis, and expression of mutants in yeast cells and trypanosomes.
