Abstract

Vitamin C is an essential micronutrient and acts both as a cofactor and as an antioxidant. Recently two rat Na+-independent ascorbic acid transporters have been cloned representing a new transporter family. We have now isolated the human homologues of the rat genes. The aims of the proposal are: 1) to elucidate the tissue and subcellular distribution of the ascorbate transporters using antibodies; 2) to study the possible differential regulation of the two human ascorbate transporters; and 3) to investigate the molecular determinants of the permeant binding site. A better definition of vitamin C transport will help in the utilisation of vitamin C supplements and in our understanding of the mechanism of transport by this new transporter family.
