Abstract

The research will enhance understanding of mechanisms for translocating nucleus- encoded proteins into chloroplasts, building on the findings of earlier work (Science, 1998, 282: 100- 103). We will identify new translocon components and elucidate the functionality of previously identified proteins. Two complementary strategies will be employed: (1) Import- defective mutants will be identified by their inability to translocate a chloroplast-specific, negatively-selectable marker. We are collaborating with researchers at the John Innes Centre on this, and have so far identified 3 putative import mutants. Cloning of two novel loci will be attempted. (2) Suppressor mutants able to translocate a chloroplast-specific, positively- selectable marker with an import-defective transit peptide will be isolated. Substantial preliminary experimentation will be necessary in this case.
