Abstract

The overall aim is to unravel the role played by FtsY in the insertion of hydrophobic proteins into the chloroplast thylakoid membrane. FtsY plays a pivotal role in bacterial membrane protein biogenesis and a chloroplast FtsY homologue has recently been cloned. We propose to capitalise on a major new development: the establishment of the first full reconstitution assay for FtsY-dependent insertion. This assay will be used to (a) establish the 'global' role of FtsY in chloroplasts, (b) elucidate the entire sequence of events in the signal recognition particle/FtsY-dependent insertion pathway and (c) determine in detail for the first time how FtsY facilitates protein insertion into the membrane-bound translocation apparatus.
