Abstract

The 3-dimensional structure of the multi-centre redox enzyme Flavocytochrome c3 is now known to a resolution of 1.8 angstrom. We shall use protein film voltammetry (PFV) to probe both intramolecular electron transfer and substrate reduction in this enzyme. PFV provides a detailed picture of the redox chemistry in both the time and potential domains; integrating thermodynamics and kinetics in a single set of experiments. A unique aspect of the work that we will be able to probe the chemistry that occurs at the FAD during turnover. Any interaction that perturbs the reduction potential of the FAD or gates electron transfer through the active site will be revealed, and the redox-state dependence and timescale of the processes will be measured.
