Abstract
We have discovered that extracellular matrix (ECM) modifies apoptosis via effects on insulin signalling in mouse mammary epithelial cells (MEC). However, IGFs are more likely to be the physiological regulators of mammary apoptosis and we hypothesise that this will depend on interplay between IGFs and ECM. To address this we will: (1) compare how IGFs and insulin regulate signal transduction and survival; (2) determine how IGF binding proteins (IGFBPs) modulate this process; (3) investigate if IGFs regulate integrins in MEC; (4) explore how ECM affects (a) IGF receptor mediated signalling, (b) the phenotypic response of MEC to IGFs and (c) IGFBP actions. Ultimately this work will elucidate the cellular mechanisms controlling mammary gland physiology.
