Abstract
The ultimate objective of this work programme is to solve the structure of photosystem two (PSII) at atomic resolution in order to further our understanding of the mechanisms of its function, especially its ability to use light energy to oxidise water and generate molecular oxygen. Two forms of 2D crystals will be analysed by electron crystallography consisting of the CP47-RC subcomplex and the oxygen evolving core dimer complex. In the case of the former, the present 8 3D map will be improved with the view to obtain 3-4 resolution. The possibility of reconstituting the four atom manganese cluster into the crystals will be explored and if successful, structurally investigated. A 3D structure of the PSII core dimer will also be elucidated at the highest possible resolution with the main aim of identifying the location of the manganese cluster relative to various protein subunits.
