Abstract

A systematic programme will investigate the molecular interactions leading to the formation of circular membrane protein assemblies. This necessitates an examination of the interactions between membrane-spanning and extrinsic domains of LH2 subunits, which determine the aggregation state and the shape of the complex. This work builds upon our recent success in determining the projection structure of the R. sphaeroides LH2 complex by electron cryomicroscopy. Three aspects of LH2 will be investigated: (1) cofactor binding (2) extrinsic interactions between the N-and C-terminal regions of the LH2 alpha and beta sub-units, and (3) the transmembrane regions of the alpha-subunits. A versatile and comprehensive genetic system, coupled with on-site expertise in spectroscopy and 2D cryo-electron microscopy will be utilised to fulfil the aims of this project.
