Abstract
Apoptosing cells undergo remarkable functional changes prior to phagocytosis by neighbouring cells, including alterations in organelle structure and function. One possibility is that compromising the specificity of membrane fusion coupled with changes in organelle motility lead to the widespread fusion of organelles which normally remain distinct, and organelles with the cell surface. We will use video microscopy in combination with organelle-specific green fluorescent protein markers to determine whether such an inappropriate fusion does occur, and the mechanisms involved. In addition, by combing high resolution differential interference contrast or phase contrast microscopy with fluorescence microscopy of living cells, we will be able to order specific changes in membrane structure in relation to the timing of other cellular events.
