Abstract
The translation initiation factor eIF4F consists of the protein eIF4E, eIF4A and eIF4G. This complex has a critical role in the initiation of cellular cap-dependent protein synthesis. It has also been established that eIF4A and a region of eIF4G are required for translation of picornavirus RNA by a cap-independent mechanism. It has previously been shown that eIF4G is cleaved by the foot-and-mouth disease virus (FMDV) Leader protease. Our new data indicate that eIF4G is also cleaved by the viral 3C protease at distinct sites in vivo. Furthermore the 3C protease also induces the cleavage of eIF4A. We propose to precisely define the novel modifications of these two initiation factors and to determine the effects of the cleavage events on their properties and their roles in cap-dependent and FMDV-RNA directed translation.
