Abstract
Nano-electrospray mass spectrometry is a powerful technique currently being used by laboratories to analyse protein complexes, having advantages of specificity, speed and sensitivity over conventional techniques. The relationship between wnt and APC signalling is not known. We will identify protein signalling complex components and post-translational modifications by applying and optimising mass spectrometric analysis. Specifically, we will use this, and traditional immunoblotting procedures to analyse protein complexes of beta- catenin and adenomatous polyposis coli (APC) in different cell lines a) in which the wnt signalling pathway is active and b) in which the wnt signalling pathway is not stimulated.
