Abstract

A more complete understanding of the molecular regulation of preadipocyte differentiation may lead to novel strategies for controlling obesity. During the differentiation of 3T3-F442A preadipocytes, a well-defined model of mammalian preadipocyte differentiation, we have found that certain isoforms of protein kinase C (PKC) are required at distinct stages of the process. This project aims to determine 1) the changes in the subcellular distribution patterns and catalytic activities of PKC isoforms during preadipocyte differentiation, 2) the molecular basis for the regulation of clonal expansion of immature adipocytes by PKC-gamma and 3) the molecular basis for regulation of terminal differentiation by PKC-epsilon.
